Son of Meth­od­ist preach­er Lu­ther J. Cox and bro­ther of Christ­o­pher Cox, Sam­u­el was a first cou­sin to Bi­shop John C. Keen­er. In 1844, he joined the Meth­od­ist Mar­y­land Con­fer­ence, of which his father had been one of the or­gan­iz­ers in 1828. Af­ter pas­tor­ates in Wash­ing­ton, DC, and else­where, in 1853 he be­came Pro­fess­or of Men­tal and Mor­al Phil­o­so­phy at Ma­di­son Coll­ege, Un­ion­town, Penn­syl­van­ia. He held that po­si­tion a num­ber of years, then en­gaged in edu­ca­tion­al work in Vir­gin­ia and Al­a­ba­ma un­til 1866, when he joined the Meth­od­ist Epis­co­pal Church, South. He then served as an ed­u­cat­or and pas­tor in Bal­ti­more, Mar­y­land, Wash­ing­ton, DC and else­where; ed­it­or of the Epis­co­pal Meth­od­ist, the Bal­ti­more Chris­tian Ad­vo­cate, and the Bal­ti­more and Rich­mond Chris­tian Ad­vo­cate; and mem­ber of the com­mit­tee of nine which com­piled the 1886-88 hym­nal of the Meth­od­ist Epis­co­pal Church, South.
